V 1. polovici 14. storo\u010dia bola postaven\u00e1 na ju\u017enom okraji Pre\u0161ova budova mestsk\u00e9ho \u0161pit\u00e1la, ku ktorej bola e\u0161te v tom istom storo\u010d\u00ed pristaven\u00e1 men\u0161ia kaplnka. Na jej mieste za\u010dali stava\u0165 roku 1429 nov\u00fd \u0161pit\u00e1lsky kostol, zasv\u00e4ten\u00fd Panne M\u00e1rii. V jeho susedstve sa nach\u00e1dzala nemocnica a chudobinec. Po n\u00e1stupe reform\u00e1cie sa \u0161pit\u00e1lsky kostol stal evanjelick\u00fdm kostolom slovensk\u00e9ho obyvate\u013estva Pre\u0161ova. Po nariaden\u00ed panovn\u00edka Leopolda I. bol v roku 1673 kostol odovzdan\u00fd reholi minoritov, ktor\u00ed pri\u0161li do Pre\u0161ova v tom istom roku. Okrem kostola minoriti dostali do vlastn\u00edctva aj nemocnicu \u2013 \u0161pit\u00e1l. V 17. st. bol spojen\u00fd s minoritsk\u00fdm kl\u00e1\u0161torom a v 18. storo\u010d\u00ed barokovo prestavan\u00fd do s\u00fa\u010dasnej podoby. V roku 1757 vznikli \u0161tyri v\u00e4\u010d\u0161ie fresky zo \u017eivota sv. J\u00e1na Krstite\u013ea. Dodnes s\u00fa zachovan\u00e9 na strope chr\u00e1movej lode. Kostol patril od r. 1818 gr\u00e9ckokatol\u00edckemu biskupstvu. V severnej kaplnke je pred olt\u00e1rom umiestnen\u00fd sarkof\u00e1g s pozostatkami biskupa Petra Pavla Gojdi\u010da. Jeho pozostatky do sarkof\u00e1gu ulo\u017eili po jeho blahore\u010den\u00ed v roku 2001. Na severnej stene katedr\u00e1ly je za sklom upevnen\u00e1 k\u00f3pia Tur\u00ednskeho pl\u00e1tna, ktor\u00e9 pre\u0161ovsk\u00e1 katedr\u00e1la dostala ako dar od Tur\u00ednskeho arcibiskupstva v Taliansku v roku 2003. Ide o faksimile, ktor\u00e9 bolo vyroben\u00e9 len v \u0161tyroch exempl\u00e1roch. V 50. rokoch bol chr\u00e1m dan\u00fd do u\u017e\u00edvania pravosl\u00e1vnej cirkvi, ke\u010f bola v \u010ceskoslovensku zru\u0161en\u00e1 gr\u00e9ckokatol\u00edcka cirkev.
